Do I have to count note by note, or can I type one total?

Either. Tapping through the note tiles is quicker than it sounds and it catches your own miscounts. If you have already counted on paper, put the whole amount in the coins tile instead and be done. The POS stores what you counted and the difference against what it expected, so both routes end in the same place.
